In order to determine the suitability of thecyprinodont fish Aphanius dispar as a biologicalcontrol agent for mosquito larvae, the effects oftemperature, salinity, feeding rate and photoperiod onits reproductive condition were studied. The factorsexamined were the gonado-somatic index (GSI) and fourdistinct stages of cell maturation in the ovaries(oogonia, primary oocytes, secondary oocytes andmature ova). The fish were found to be tolerant to awide range of the examined factors. GSI's were notaffected by ambient temperatures over the range of18 °C to 37 °C. However, oocyte maturationstages increased as temperatures rose from 18 °Cto 27 °C, with mature ova first being found at thelatter temperature. No significant differences amongmaturation stages were found between27 °C–37 °C. At salinities ranging from of0 ppt to 56 ppt, no significant differences were found betweenmean GSI's, except for the two extremes of the testrange. The ovaries of fish kept in distilled watercontained primordial germ cells only. In feedingexperiments, significant differences were found onlybetween the ovaries (GSI's and oocyte maturationstages) of fish deprived of food and those fed at1% body weight per day. Regarding photoperiod,a decrease in day length from 14L:10D (the longest dayof the year) to 10L:14D (the shortest) caused adecrease in oocyte maturation stages. Fish at theintermediate level (12L:12D) still had ovariessuitable for spawning, whereas those at 10L:14D didnot. The results of this study show that limitinglevels for adequate reproductive condition were foundfor some of the factors investigated, however, rangesof the factors above these levels were relativelylarge. That Aphanius dispar's reproductivecondition may be controlled in an artificialenvironment is essential for it to be mass produced,and a prerequisite for its use in the biologicalcontrol of mosquito larvae.  相似文献

Summary Intratumor injections of the aqueous phase of phenol-water extracts of Re mutant Salmonella typhimurium (Re glycolipid) in combination with trehalose dimycolate at dose levels of 150 to 15 g were consistently and highly effective (65–93%) in producing regression of line-10 tumors in strain-2 guinea pigs. We observed that the rate of regression was more rapid than that seen after treatment with cell walls from Mycobacterium bovis strain Bacillus of Calmette and Guèrin (BCG). Arabinose mycolate could be substituted for trehalose dimycolate in the Re glycolipid-mycolate mixture without appreciably compromising antitumor activity, providing that the level of arabinose mycolate was not reduced below 15 g. In addition to the Re glycolipid preparation, similarly prepared aqueous extracts from Mycobacterium bovis strain BCG and strain AN5 in combination with trehalose dimycolate also possessed tumor-regressive activity. The activity of these last extracts was reduced when the arabinose mycolate was substituted for the trehalose dimycolate. The aqueous extract of a rickettsia, Coxiella burnetii, in combination with either trehalose dimycolate or arabinose mycolate was also active (50 and 80% tumor regression rates, respectively). Intracutaneous administration of Re glycolipid or aqueous extracts from BCG in combination with trehalose or arabinose mycolates did not produce life-threatening, clinical signs of toxicity in young mice. If additional toxicity studies demonstrate that adverse side effects can be satisfactorily controlled, these watersoluble extracts may prove beneficial in the treatment of spontaneous tumors of humans and other animals.  相似文献

Summary Cell free extracts from a streptomycin-resistant E. coli mutant which is also temperature-sensitive for Q phage were studied for suppression of a nonsense mutation at various temperatures. The streptomycin-resistant ribosomes of the mutant were found to be temperature-sensitive in suppression of an amber mutation in f2 phage coat protein while retaining the ability to synthesize proteins at an elevated temperature (42° C). The restriction of amber suppression at 42° C is assumed to be related to an alteration in the ribosomal protein S12 of the streptomycin-resistant mutant which also causes a change in its electrophoretic mobility.  相似文献

Cycloheximide inhibits polar indoleacetic acid transport in midrib tissues of leaves of citrus (Citrus sinensis [L.] Osbeck) and poplar (Populus deltoides Bartr.) as measured by the donor-receiver agar cylinder technique. It appears that the mechanism of auxin transport inhibition by cycloheximide consists in arresting protein synthesis and not in the disruption of energy flow. The interpretation of the data takes into account the involvement of either a carrier protein or auxin-induced proton excretion in auxin transport.  相似文献

The relationship between the relative abundance of ureides ([ureide-N/ureide-N plus nitrate-N] × 100) in the shoot axis (stems plus petioles), nodulated roots and leaflets of “Bragg” soybean (Glycine max [L.] Merrill) and the symbiotic dependence of these plants was examined under glass-house conditions. Plants, inoculated with effective Rhizobium japonicum CB1809, were grown with their roots exposed continuously to a nutrient solution containing either 0, 1.5, 3.0, 6.0 or 12.0 millimolar NO3-N per liter. Nodulation and N2-acetylene fixation were correlated inversely with the level of nitrate. Seasonal acetylene reduction profiles for each of the nitrate treatments were integrated and the symbiotic dependence ([N2 fixed per total plant N] × 100) determined using a conversion ratio of 1.5:1 (acetylene reduced:N2 fixed), calculated from the zero NO3 treatment. Examination of the nitrogenous solutes of the shoot axis and nodulated roots showed linear relationships between the relative abundance of ureides and the symbiotic dependence of the plants. Two standard curves, depicting these relationships during vegetative and reproductive growth, were drawn for each plant part. The overriding effect of plant age invalidated any attempt to develop a standard relationship for leaflets. Data from two diurnal studies suggested that relative ureides were insensitive to diurnal fluctuations, thus simplifying sampling procedures. Plant material could be stored at ambient temperatures (20-30°C) for up to 24 h without affecting the relative concentration of ureides and nitrate. It is suggested that the shoot axis provides the most suitable target organ when using this technique as a quantitative assay for N2 fixation because of ease of sampling of these tissues, especially with field-grown plants.  相似文献

When mature green harvested Shamouti oranges (Citrus sinensis L. Osbeck) were exposed to 35 μl/liter of ethylene, a 3-fold increase in free abscisic acid (ABA) of the flavedo could be detected after 12 hours and a 10-fold increase after 24 hours, while chlorophyll destruction did not exceed 20%. The increase in free ABA continued up to 24 hours and leveled off. Bound ABA accumulated strongly after 24 hours suggesting that excess of free ABA was being converted into the bound form. Similar increases in ABA upon exposure to ethylene occurred also in fully mature orange fruits which had already lost all of their chlorophyll, in white and green portions of the flavedo of variegated lemons, and in the colorless albedo of Eureka lemons.  相似文献

Astroglia are neural cells, heterogeneous in form and function, which act as supportive elements of the central nervous system; astrocytes contribute to all aspects of neural functions in health and disease. Through their highly ramified processes, astrocytes form close physical contacts with synapses and blood vessels, and are integrated into functional syncytia by gap junctions. Astrocytes interact among themselves and with other cells types (e.g., neurons, microglia, blood vessel cells) by an elaborate repertoire of chemical messengers and receptors; astrocytes also influence neural plasticity and synaptic transmission through maintaining homeostasis of neurotransmitters, K+ buffering, synaptic isolation and control over synaptogenesis and synaptic elimination. Satellite glial cells (SGCs) are the most abundant glial cells in sensory ganglia, and are believed to play major roles in sensory functions, but so far research into SGCs attracted relatively little attention. In this review we compare SGCs to astrocytes with the purpose of using the vast knowledge on astrocytes to explore new aspects of SGCs. We survey the main properties of these two cells types and highlight similarities and differences between them. We conclude that despite the much greater diversity in morphology and signaling mechanisms of astrocytes, there are some parallels between them and SGCs. Both types serve as boundary cells, separating different compartments in the nervous system, but much more needs to be learned on this aspect of SGCs. Astrocytes and SGCs employ chemical messengers and calcium waves for intercellular signaling, but their significance is still poorly understood for both cell types. Both types undergo major changes under pathological conditions, which have a protective function, but an also contribute to disease, and chronic pain in particular. The knowledge obtained on astrocytes is likely to benefit future research on SGCs.
